Well.... study of gene expression can have many ways.... 
1. one common way scientist do is to introduce mutation in organism or plant... so it can test the function of the gene. 
2. Also.... if you are studing cellular biology or genetics... you can alter the mRNA translation process.... so it result the alteration of gene function, like alter the operon, the promoter or the translator. 
3. Also, if you want to know the allele of certain trait... you can cross your unknow with the know recessive trait to see if your trait is homozygous or heterozygous....  
4. One last method is to use plasmid vector, like you use E.coli to cut human genome and you insert this cut human genome into E.colid plasmid and grow in cell culture to see if there is any different to the normal E.coli.....
